چکیده

An increase in the intracellular Ca” concentration of human erythrocytes results in profound morphological changes and loss of plasma membrane in the form of small vesicles [ 1,2]. It is of interest to determine whether this behaviour could be related to a Ca2+-induced instability in the bilayer structure of the membrane lipid. We have therefore examined the influence of Ca2+ on model membrane systems with lipid compositions approximating those of the outer and inner monolayer of the erythrocyte membrane. We show that whereas the outer monolayer lipids are unaffected by Ca2+, a large proportion of the inner monolayer lipids adopt the hexagonal (H,) phase in the presence of Cap. We suggest that this tendency of the inner monolayer to adopt non-bilayer configurations could be directly related to the fusion event involved in vesicle ejection following CaZ* influx.

The anticancer drug cisplatin interacts with the human erythrocyte membrane.

Drugs which exert their effects by interacting with DNA cause structural and functional membrane alterations which may be essential for growth inhibition by these agents. This paper describes the interaction of cisplatin with the human erythrocyte membrane and models constituted by bilayers of dimyristoylphosphatidylethanolamine (DMPE) and diacylphosphatidylserine (DAPS), representative of phos...

Behenic Acid Monolayer and Bilayer Assemblies- A Study of Concanavalin A (Con A) Adsorption and its Interaction with Dextran Using Surface Plasmon Resonance Spectroscopy and Microscopy

Deposition of behenic acid (B.A) mono and bilayers onto gold coated surfaces was performed by Langmuir-Blodget dip-casting technique. Surface Plasmon resonance (SPR) and ellipsometry methods were employed for investigation of the monolayer and bilayer films. The adsorption of the biologically important molecule Concanavalin A (Con A) from bulk solution to these monolayers and bilayers as we...

Molecular Insight into the Mutual Interactions of Two Transmembrane Domains of Human Glycine Receptor (TM23-GlyR), with the Lipid Bilayers

Appearing as a computational microscope, MD simulation can ‘zoom in’ to atomic resolution to assess detailed interactions of a membrane protein with its surrounding lipids, which play important roles in the stability and function of such proteins. This study has employed the molecular dynamics (MD) simulations, to determine the effect of added DMPC or DMTAP molecules on the structure of D...

The anticancer drug chlorambucil interacts with the human erythrocyte membrane and model phospholipid bilayers.

The plasma membrane has gained increasing attention as a possible target of antitumor drugs. It has been reported that they act as growth factor antagonists, growth factor receptor blockers, interfere with mitogenic signal transduction or exert direct cytotoxic effects. Chlorambucil (4-[p-(bis[2-chloroethyl]amino)phenyl]butyric acid) is an alkylating agent widely used in the treatment of chroni...

Lateral tensions and pressures in membranes and lipid monolayers.

The effects of lateral tension on the properties of membranes are often explained in comparison with analogous experiments on monolayers, which yield more detailed data. To calculate the effects of changes in tension on the composition of, or incorporation of amphiphiles into membranes we examine (i) the fidelity of the monolayer analogy, (ii) the range of possible tensions in a membrane, and t...
